BASIC TECHNIQUE OF MICROPROPAGATION
Before we start, I just want to explain about what is micropropagation?
Micropropagation is an advanced vegetative propagation technology for producing a large number of genetically superior and pathogen-free transplants in a limited time and space.
Well let's start with first step.
#STEP 1 SELECTION OF MOTHER PLANT
- The selected mother plants must be healthy and free from pest and disease.
#STEP 2 PREPARATION OF EXPLANT MATERIALS
- Explants need to be surface sterilized (any part of the vegetative parts; shoot )
#STEP 3 INITIATION OF SHOOTS
- Mass formation of adventitious shoot around the explant
#STEP 4 SUBCULTURE
- Shoots produced from the initial culture are isolated and onto suitable medium. Normally, cytokinin proportion is increased to produce numerous shoots
#STEP 5 SHOOT INDUCTION
- Shoot are seperated manually from clusters and induced to a certain height.
#STEP 6 ROOT INDUCTION
- Each individual plantlets are transferred on a rooting medium containing auxin.
#STEP 7 ACCLIMATIZATION
- The process is important in which an individual plantlets adapt to a gradual change in its environment (such as a change in temperature, humidity, photoperiod, or pH) , allowing it to maintain performace (survive) across a range.
#STEP 8 MANAGEMENT OF SECONDARY HARDENING
- Plantlet in poly-bag are placed on benches or raised beds under 70% shades
So after we learned about the basic technique of micropropagation, we had chance to learned how to planting the banana trees.
